چکیده
The transcriptional co-activator PGC-1 and the NAD -dependent deacetylase SIRT1 are considered important inducers of mitochondrial biogenesis because in the nucleus they regulate transcription of nucleus-encodedmitochondrial genes. We demonstrate that PGC-1 and SIRT1 are also present inside mitochondria and are in close proximity tomtDNA. They interact with mitochondrial transcription factor A (TFAM) as assessed by confocal microscopy analysis and by blue nativePAGE. Nucleoid purification allowed us to identify SIRT1 and PGC-1 as proteins associated with native and cross-linked nucleoids, respectively. After mtDNA immunoprecipitation analysis, carried out on mitochondrial extracts, we found that PGC-1 is present on the same D-loop region recognized by TFAM. Finally, by oligonucleotide pulldown assay, we found PGC-1 and SIRT1 associated with the TFAM consensus sequence (human mitochondrial transcription factor-binding site H). The results obtained suggest that in mitochondria PGC-1 and SIRT1 may function as their nuclear counterparts and represent the genuine factors mediating the cross-talk between nuclear and mitochondrial genome. Finally, this work adds new knowledge on the function of SIRT1 and PGC-1 and highlights the direct involvement of such proteins in regulation of mitochondrial biogenesis.

Sirtuin 1 activation enhances the PGC-1α/mitochondrial antioxidant system pathway in status epilepticus.
Sirtuin 1 (SIRT1) regulates numerous neuronal processes, including metabolism, antioxidation and aging, through activation of peroxisome proliferator-activated receptor coactivator 1-α (PGC-1α), an upstream regulator of mitochondrial biogenesis and function. However, the role of SIRT1 in the oxidative stress induced by seizures has yet to be elucidated. The present study aimed to investigate wh...

Silent mating type information regulation 2 homolog 1 (SIRT1) is implicated in the control of skeletal muscle mitochondrial content and function through deacetylation of peroxisome proliferator-activated receptor γ coactivator-1α (PGC-1α) and participation in the SIRT1/PGC-1α axis. The SIRT1/PGC-1α axis control of skeletal muscle mitochondrial biogenesis is an important therapeutic target for o...

BACKGROUND Abnormalities of brain energy metabolism are involved in Alzheimer disease (AD). Sirtuin 1 (SIRT1) is a class III histone deacetylase and activates peroxisome proliferator-activated receptor-γ co-activator-1α (PGC-1α), which enhances mitochondrial biogenesis and energy homeostasis. Electroacupuncture (EA) has been reported to improve brain energy metabolism in AD. However, the effect...

IN 2007, Gerhart-Hines et al. (12) published a series of cell culture experiments that implicated Sirtuin-1 (SIRT1) in the regulation of mitochondrial content and fatty acid oxidation in skeletal muscle. Specifically, this study proposed that SIRT1 deacetylates peroxisome proliferator-activated receptor coactivator (PGC)-1 , resulting in increased transcription of genes associated with mitochon...
